Assay specificity The assay is specific for pathogenic Cryptosporidium species and Giardia assemblages A&B. Other related protozoa that were tested (e.g., Entamoeba histolitica, Babesia canis, Babesia gibsoni, Theileria annae, Toxoplasma gondii and Trypanosoma brucei) showed negative results under the same conditions. CG_Graph Assay Quantification The above is a typical standard plot of serial decimal dilutions of target Cryptosporidium and Giardia DNA sequences (green dots). Blue dots indicate water samples containing 30 and 3 Cysts/Oocysts, respectively. Environmental water samples were spiked with Cysts and Oocysts from validated spiking stocks (Waterborne, Inc.) and tested with AquatiqPCR©-C/G kit. Results The number of Giardia Cysts and Cryptosporidium Oocytes can be deduced by a standard plot based on serial decimal dilutions of target DNA oligo’s. Taking into account that every Giardia Cyst contains 8 sets of genomic DNA and each Cryptosporidium Oocyst contains 4 sets, we are able to translate the DNA copy number from the qPCR reaction into actual numbers of Cyst/Oocyte.

AquatiqPCR©-C/G vs. USEPA Method 1623 in environmental water filtrate. A side-by-side study was conducted at the Central Lab of Mekorot Ltd. (Israel National Water Company). Filtrates of 50 liter environmental water samples were spiked with 5 or 10 Giardia cysts and 5 or 10 Cryptosporidium oocytes, respectively. Results Numbers above the columns indicates the ΔCq values as per the qPCR analysis using AquatiqPCR©-C/G kit. Numbers underneath the columns shows the results of USEPA method 1623 (number of oocysts/cysts after extraction, immunostaining and microscopic examination). AquatiqPCR©-C/G kit was able to detected Cryptosporidium and Giardia DNA in all samples tested whereas the microscopic examination was inaccurate and failed to detect Cryptosporidium in low levels (false negative).
